产品介绍 HOXB4 is a homeodomain-containing transcription factor with diverse roles in embryonic development and the regulation of adult stem cells. It promotes proliferation of hematopoietic stem cells and can both activate and repress apoptosis.

OXB4通过激活不同的信号通路增强HSC的自我更新,同时又不影响维持正常稳态造血的调控机制。提高HOXB4的表达水平,能够极大地增强HSC的自我更新功能,但基本不影响细胞的分化、系特异性及终末细胞的形态和功能。另外，HOXB4还可增强胚胎干细胞(embryonic stem cell,ESC)的造血潜能,促进ESC向造血细胞分化。

Paraformaldehyde-fixed, paraffin embedded (Mouse placenta); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (HOXB4) Polyclonal Antibody, Unconjugated (bs-3650R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.Blank control:K562.
Primary Antibody (green line): Rabbit Anti-HOXB4 antibody (bs-3650R)
Dilution: 2μg /10^6 cells;
Isotype Control Antibody (orange line): Rabbit IgG .
Secondary Antibody : Goat anti-rabbit IgG-PE

Protocol
The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 90% ice-cold methanol for 20 min at-20℃. The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.Blank control: K562.
